Curtin University adjunct research fellow Gautam Sethi says most diseases, including cancer, are caused by the deregulation of multiple genes. Curcumin safe in high doses
A/Prof Sethi says curcumin is possibly the only drug that can be given at high dosesup to 12gwithout any toxicity. It can target most of the oncogenic proteins like NF-kB, STAT3, AP-1, he says.
A/Prof Sethi says the only known side effect of the agent is blood thinning, and therefore advises against taking curcumin if undergoing surgery. A/Prof Sethi says it would be ideal to combine curcumin with other drugs or natural compounds, like piperine, an alkaloid found in pepper to increase its bioavailabilty.
If we combine it with piperine we see viability increase by 2000 per cent 45 minutes after administering the curcumin, he says. A/Prof Sethi says there is a lack of data to explain the underlying mechanism of its effect, however, it is known for its anti-inflammatory effects. It has been shown that most chronic diseases, including cancer, are caused by inflammation and can be treated by anti-inflammatory agents.
He says more work needs to be done to improve curcumins viability, as body tissues quickly absorb it.

Multiple Myeloma
The chemopreventive potential of curcumin in monoclonal gammopathy of undetermined significance (MGUS), a high-risk condition for progression to multiple myeloma, was tested by Golombick et al., in a cross-over study design [306]. Out of 27 patients, 17 received 4000 mg of curcumin for 3 months before cross-over to placebo. The rest received placebo first, followed by curcumin. The levels of paraprotein and urinary N-telopeptide from type I collagen decreased in some patients. The same group reported beneficial effects of curcumin in MGUS in another randomized, double blind, placebo-controlled cross-over study [314]. A phase I/II trial by Vadhan-Raj et al., in 29 patients with asymptomatic, relapsed, or plateau phase multiple myeloma showed a decrease in expression of NF-κB, COX-2, and STAT3 in peripheral blood mononuclear cells and stable maintenance of disease [304]. The patients were given either oral dose of curcumin alone (2000, 4000, 6000, 8000, or 12,000 mg/day) or in combination with 10 mg of bioperine. 12 patients continued the treatment for 12 weeks, followed by combination treatment for five patients (one at a dose of 4000, two at 6000 mg, and two at 8000 mg) for 1 year. Further, well-controlled clinical trials with larger sample sizes are required to substantiate the efficacy of curcumin against multiple myeloma.
Pancreatic Cancer
Dhillon et al., conducted a phase II clinical trial in 25 advanced pancreatic cancer patients [305]. An oral dose of 8000 mg of curcumin was given every day until disease progression, with restaging performed every 2 months. Low, steady-state levels of curcumin and its conjugates were detected in the peripheral circulation for the first 4 weeks with a peak at 2242 ng/mL. In two patients, a biological effect was seen and in one the disease was maintained stably for 18 months. Remarkably, a significant but brief tumor regression was seen in one patient with a concomitant increase in serum cytokine levels. NF-κB, COX-2, and STAT3 suppression in peripheral mononuclear cells was also observed in the patients. Although oral curcumin was found to be safe and well-tolerated, poor bioavailability still remains an issue, which partly explains the biologic activity observed only in some patients. In another open-label, phase II trial, a combination of curcumin and gemcitabine was tested in advanced pancreatic cancer patients [308]. Seventeen patients were administered 8000 mg of curcumin orally every day for 4 weeks while 1000 mg/m3 of gemcitabine were given intravenously three times a week. In five patients, curcumin or the whole treatment was discontinued due to toxicity and one patient died suddenly. In the remaining 11 patients, a partial response was seen; four had stable disease while in six the tumor progressed. Tumor progression time was 112 months, with overall survival time of 124 months, indicating a modest efficacy of the combination therapy. Also, the authors concluded that 8000 mg/day of curcumin with gemcitabine was above the maximum tolerated dose. In a recent phase I/II trial by Kanai et al., a similar combination of gemcitabine and curcumin was used to treat gemcitabine-resistant pancreatic cancer in 21 patients [311]. In contrast, the combination of gemcitabine and curcumin 8000 mg/day was found to be safe and well tolerated in this study. Studies in a larger cohort are required to validate the results.
Researchers Conclusion
A plethora of in vitro and in vivo research together with clinical trials conducted over the past few decades substantiate the potential of curcumin as an anti-cancer agent. At the molecular level, curcumin targets numerous pathways, highlighting its ability to inhibit carcinogenesis at multiple levels and thus, potentially circumventing the development of resistance. However, there is a paucity of data to explain the underlying mechanism of its activity. Clinical trials with curcumin indicate safety, tolerability, non-toxicity (even up to doses of 8000 mg/day), and efficacy. These studies provide a solid foundation for more well-controlled studies in larger cohorts as well as open avenues for future drug development. However, curcumin activity is limited by its poor bioavailability and some possible adverse effects. The development of formulations of curcumin in the form of nanoparticles, liposomes, micelles or phospholipid complexes to enhance its bioavailability and efficacy are still in its early stages. Nonetheless, curcumin has established itself as a safe and promising molecule for the prevention and therapy of not only cancer but also other inflammation-driven diseases.

Curcumin-based anti-prostate cancer agents are being developed on the premise that it acts as a multitarget drug and has the potential to treat castration-resistant prostate cancer.
The study reports that out of the three curcuminoids, demethoxycurcumin shows the most efficient anti-tumor activity against prostate cancer cells.
Curcumin acts on many biochemical pathways in order to prevent cell proliferation and also to cause cancer cell death.
For example, protein kinase D1 is a biological molecule that is expressed in high amounts in normal prostate cells but is reduced in prostate cancer cells.
Curcumin activates this protein and thereby prevents growth and spread of prostate cancer.

Tumor necrosis factor is perhaps the principal protein involved in inflammation. Additionally, modulation of its activity can induce cell death.
TRAIL-mediated immunotherapy works on the principle of activating this protein to destroy cancerous cells. But cancer cells develop a resistance towards it.
Research shows that curcumin reverses this resistance and sensitizes prostate cancer cell to this treatment.
Curcumin also increases the efficacy of first-line treatment drugs like bicalutamide and docetaxel which are used to treat prostate cancer.

Prostatitis is the inflammation of the prostate gland. It can serve as a risk factor for prostate cancer.
As mentioned earlier, curcumin displays its chemopreventive potential by reducing inflammation in the prostate gland.

A study was conducted where curcumin along with other herbal extracts was evaluated for their effect on bacterial prostatitis.

A study published in the Journal of Medicinal Food titled, “Efficacy and Safety of a Flaxseed Hull Extract in the Symptomatic Management of Benign Prostatic Hyperplasia: A Parallel, Randomized, Double-Blind, Placebo-Controlled, Pilot Study,” reveals that men who consumed an extract of flaxseed for swollen prostate (i.e. benign prostatic hyperplasia) saw significant improvement.
According to the study, The American Urology Association (AUA) Guideline, 2010, defines benign prostatic hyperplasia (BPH) as proliferation of smooth muscles and epithelial cells within the prostatic transition zone. The typical symptoms include:

Decreased and intermittent force of stream
Sensation of incomplete bladder emptying.
BPH is one of the most widespread health conditions faced by men today, with the incidence rising to nearly 50% of the male population by the eighth decade of their lives. Current approaches include side effect heavy pharmaceuticals such as alpha adrenergic blockers and 5 a-reductase inhibitors, as well “gold standard” surgical procedures such as transurethral resection of the prostate (TURP), which so consistently produces surgery related impairments that there is an entire syndrome named after these side effects, namely, Transurethral Resection of the Prostate (TURP) Syndrome.
The human cost in both financial and existential terms is immense. This is why interest in safe, natural, affordable and effective alternatives is growing, and why the researchers felt compelled to test the hypothesis that eating flaxseed might provide an ideal alternative approach.
In the randomized, double-blinded, and placebo-controlled study, three groups of men, aged 45-75, with newly diagnosed BPH and an American Urological Association Symptom Index (AUASI) score of 13 , were given either a placebo, or a low or high dose of a lignin-rich flaxseed hull extract. More specifically:
“Study treatment consisted of 500 or 1000 mg of extract containing 100 mg (low-dose active [LDA] group, n = 26) or 200 mg (high-dose active [HDA] group, n = 26) of secoisolariciresinol diglucoside (SDG), respec- tively. The placebo (P) group (n = 28) received matching maltodextrin capsules. Sixty subjects (LDA [n = 19], HDA [n = 20], and P [n = 21]) completed the study as per the protocol requirements. Change in the AUASI score within a period of 8 weeks, from baseline to end of treatment, was assessed.”
The promising results were reported as follows:
“In this study, supplementation with the flaxseed hull extract provided greater relief than placebo in obstructive symptoms of BPH, such as sensation of incomplete bladder emptying, ”stopping and starting” while urinating, weak urinary stream, and ”straining while urinating.” Low and high doses of the flaxseed extract provided statistically significant improvements in the scores of these obstructive symptoms at week 8 as compared with baseline. In contrast to this, the placebo group did not show a statistically significant improvement with respect to these obstructive symptoms.”
The treatment group did not see any sign of increased side effects relative to the placebo group, indicating how remarkably safe flaxseed is versus conventional treatment. Also, because the placebo group also saw a significant improvement, it is possible that the strong placebo effect may have overshadowed the real power of the intervention. We hope that in the future a non-pharmaceutical industry funded study compares flaxseed to both pharmaceutical and surgical intervention to assess its true power.
Another important observation is that the flaxseed groups saw an increase in free testosterone, as well as a decrease in the testosterone metabolite DHT, which is known to contribute to prostate growth both benign and malignant. The placebo group, on the other hand, saw both an increase in testosterone and DHT. This also means that aging men, whose testosterone levels often plunge in relation to their estrogen levels, might experience additional hormonal benefits from eating flaxseed that could contribute to their overall state of well-being.
Concluding Remarks
This study is highly promising for a number of reasons. First, flaxseed is a safe, time-tested, and highly nutritious food that has been researched for a wide range of health benefits. Anything that can be used as a nourishing food while at the same time significantly relieving human suffering from a common disease, is highly compelling.
But there is a deeper reason why flaxseed may work so well. Our bodies co-evolved with certain foods for hundreds of thousands of years. Certain foods contains phytocompounds which our bodies have become dependent upon for self-regulation and homeostasis It is possible that lignins are vitally important, as essential as vitamins for maintaining optimal health. Obviously BPH is not caused by a lack of surgery or synthetic chemicals, but it might be true to say that BPH is caused by the lack of an ideal diet which would include lignin rich foods like flaxseed. Cleary, therefore, flaxseed could be considered part of a root cause resolution approach to BPH, versus looking it as just a palliative “disease management” tool like most of the approaches within the conventional armamentarium. We also recently reported on a study which found, in the first study of its kind, that sesame seed contains as high — if not a higher density — of lignans than flaxseed.
Finally, BPH falls within the category of a condition that involves the abnormal growth of the very same epithelial tissue that cancer emerges within. Since flaxseed has been found to prevent and even regress cancerous epithelial growths, such as breast cancer, it is highly likely that flaxseed consumption may also be the best insurance policy out there to prevent prostate cancer from developing.
No pharmaceutical drug on the planet can do
what curcumin can do
Many of my patients ask me what the difference is between curcumin
and turmeric. Turmeric is the common name of the plant Curcuma
longa. Curcumin is the active compound within the plant that’s used
for its medicinal properties.
If you’re in a fight for your life, you need to know that not just any old
curcumin will do. And while I believe that adding it to your food is
beneficial to your health, simply cooking with it is not the best way to
maximize its anti-cancer benefits. When turmeric powder is used as a
spice, it typically contains only about 3% curcumin.
Early studies uncovered the fact that curcumin is not easily absorbed
on its own. (Scientists call that poor bioavailability.) In some studies,
patients were given as much as 8 grams (8,000 mg) of curcumin, which
can irritate the gastrointestinal system. It is not known how the results
could have been affected by using a form of curcumin that was able to
be absorbed more easily.
Apigenin is a flavonoid is an antioxidant powerhouse in the same

But one look at the evidence shows that it is only a matter of time before
this compound carves out its own place on the supplement shelves.
Scientists have discovered that apigenin launches an attack against
abnormal cells on several different fronts all of which act in tandem
to thwart cancer growth.
One of the most recent studies showed that apigenin is able to undermine
cancer cells ability to cheat death (which they do by tricking your body
natural cell cycles). This is the loophole that allows tumors to grow
unchecked. (In fact, researchers have found that cancer cells use sugar
to do this.
According to this new study, apigenin restores a normal life cycle to
cancer cells by interacting with key proteins that influence RNA. This
gives the malignant cells a different set of instructions that makes them
vulnerable to programmed cell death, when they otherwise would not be.
But its not the only trick apigenin has up its sleeve.
Cancer cells are able to avoid their natural fate by subverting a protein
called p53. This is a tumor suppressor that triggers the death of cells
with damaged DNA. Research shows that apigenin is able to activate
p53. And this lowers cancer cells resistance to chemotherapy and
improves responses to treatment.
Yet another recent study showed that apigenin also blocks vascular
endothelial growth factor (VEGF) This is the same factor that triggers
angiogenesis a technical term for the new blood vessels that tumors
grow to feed themselves.
To put it simply, apigenin starves malignant tumors. And this slams the
brakes on cancer development and growth.
And now, apigenin is unique cancer-fighting powers are at the center
of another recent study this time focusing on one of the deadliest
forms of the disease.
A team of researchers treated human pancreatic cancer cell lines with
apigenin, along with another flavonoid called luteolin, extracted from
celery and artichoke. And they found that these compounds increased
cancer cell death rates from 8.4 percent to 43.8 percent.
Their conclusion? Pretreatment with apigenin might significantly boost
the effectiveness of chemotherapy in cases of pancreatic cancer.
The researchers made a point of noting that you likely would not be able
to eat enough of either vegetable to consume the amounts of flavonoids
used in this study. And there is always the question of whether lab results
will carry over in a clinical setting.
But their hope and mine, as well is that this research will lead to
the development of new supplements to aid in the fight against cancer.
As of now, stand-alone apigenin products are scant. (Although other
sources, like chamomile extracts, are more common.) I expect that
to change in the not-so-distant future, as long as studies like this one
keep making headlines.
But until then, you can not go wrong by filling up on dietary sources of
apigenin. Eat yourself away from a dozen different cancers
Numerous studies on a variety of human cell lines show that apigenin
can help combat a long list of cancers. These include, but are not limited
to: leukemia, lung cancer, colon cancer, ovarian cancer, cervical cancer,
endometrial cancer, breast cancer, prostate cancer, skin cancer, thyroid

Apigenin-rich foods include endive, beans, broccoli, leeks, onions, basil, rosemary, oregano, thyme, tomatoes, and especially chamomile, celery, and parsley just to name a few abundant sources.

A growing pile of research shows curcumin makes chemo more
effective so you need LESS of it and have a GREATER chance of
living cancer-free for life.
In fact, a team of stem cell researchers discovered that combining
curcumin and the chemo drug cisplatin KILLS 8 TIMES MORE CSCs
than chemo alone!
Researchers discovered this in an in vitro study with lung cancer stem
cells. In testing, 94% of the CSCs were found to survive a single exposure
to low-dose cisplatin. By combining the drug with curcumin, only half
survived. The other half were destroyed with just this one dose.
So far, curcumin has been found to supercharge the effects of 12 chemo
drugs ”including the colon cancer drug 5-fluorouracil (5-FU), which

Taiwan has had success with combining doxorubicin and curcumin in in vitro studies.
In animal studies and in vitro testing, curcumin reduced the amount of prostate-specific antigen (PSA) produced by prostate cancer cells, and triggers cancer cell death. In a
double-blind, placebo-controlled human study, researchers found that combining curcumin with soy isoflavones suppressed PSA production. At the start of the study, all
